Harnessing Lipid Metabolism to Uncover Tumor Vulnerabilities for Cancer Therapy

Abstract: Lung and pancreatic cancers, driven by KRAS mutations, exhibit profound metabolic adaptations that promote cancer progression and resistance to therapy. Among these adaptations, lipid metabolism plays a pivotal role in shaping tumor-stroma crosstalk, ultimately driving fibrosis, immune evasion, and metastasis. By integrating advanced tumor models with high-resolution lipidomic and transcriptomic analyses, we highlight how key lipid metabolic dysregulations in these aggressive cancers can be uncovered as tumor-specific vulnerabilities and identified as relevant cancer biomarkers to inform the development of personalized therapeutic strategies.
