En 2016, on estimait à 216 millions le nombre de cas de malaria. La plupart des cas de malaria restent cliniquement silencieux. Cependant, certaines personnes infectées développent des symptômes graves, entraînant la mort de plus de 500 000 personnes par an. Pourquoi certains patients meurent-ils de la malaria, tandis que d'autres ne présentent que des symptômes bénins ?
Afin de mieux comprendre la pathogenèse du malaria, le laboratoire de Pierre-Yves Mantel étudie la régulation des interactions hôte-parasite, en particulier les facteurs sécrétés par les globules rouges infectés et la manière dont ces facteurs régulent la réponse immunitaire.
Récemment, nos travaux se sont concentrés sur les vésicules extracellulaires (VE), qui sont de petites vésicules sécrétées par les globules rouges infectés par Plasmodium falciparum. Notre laboratoire a mis au point de nouveaux outils pour isoler les VE et nous avons décrit plusieurs cargaisons, notamment le contenu en ARN et en protéines. Nous avons montré que les VE transportent des miARN fonctionnels qui peuvent réguler la transcription dans les cellules réceptrices. Il est intéressant de noter que nous avons démontré que le miR451 transféré aux cellules endothéliales via les VE peut moduler la perméabilité de la barrière endothéliale en régulant directement l'expression génétique.
Le laboratoire cherche à identifier les principes fondamentaux qui expliquent le résultat des interactions hôte-parasite, dans le but de comprendre comment les infections parasitaires affectent la réponse immunitaire et la pathogenèse. Pour atteindre ces objectifs, nous utilisons la biologie cellulaire et moléculaire, l'immunologie et le modèle de paludisme chez les rongeurs.
