Retroviral vectors have been widely used to carry novel genes effectively into cells. They mimick a process which has been developed by Nature itself, the transport of oncogenes into cells to cause cancer. Genetherapy by retroviruses copies this approach and reverses the effect, tumor cells are reverted to become normal cells again.
Somehow, more sophisticated is the approach of using retroviral vectors, which can be turned on or off to activate the therapeutic gene so that the time point or even the dose can be modulated. This has been established by two different approaches, induction by an antibiotic or Interferon.
Retroviral vectors are also very useful for the analysis of tumor cells, their properties and how to make them normal again. Reduction of tumor growth in tumorbearing mice was detected with several transgenes. Our results clearly demonstrate that one needs two therapeutic genes in certain cancer cells to overcome their oncogenic potential and become normal again. Thus, two inhibitors or drugs are expected to be much superior than to use only one.
